|
マクロファージコロニー刺激因子(マクロファージコロニーしげきいんし、macrophage colony-stimulating factor;M-CSF)またはコロニー刺激因子1(colony stimulating factor 1;CSF1)は、造血幹細胞をマクロファージやその他の関連細胞に分化させる分泌型サイトカインである。また、真核細胞は、細胞間のウイルス感染に対抗するためにM-CSFを産生する。M-CSFは、実験的に報告されている3つのコロニー刺激因子(英語版)の内の1つである。M-CSFは、コロニー刺激因子1受容体(英語版)に結合する。また、M-CSFは胎盤の形成にも関与している可能性がある[5]。
構造
M-CSFはサイトカインの一種で、細胞のシグナル伝達に関与する小さなタンパク質である。活性型のM-CSFは、細胞外でジスルフィド結合したホモ二量体として存在しており、膜結合型前駆体のタンパク質分解により生成されると考えられている[5]。
この遺伝子には、3つの異なるアイソフォーム(プロテオグリカン、糖タンパク質、細胞表面タンパク質)[6]をコードする4つの転写バリアントが見つかっている[5]。
機能
M-CSF(またはCSF-1)は、単球、マクロファージ、および骨髄前駆細胞の増殖、分化、生存に関与する造血成長因子である[7]。M-CSFは、マクロファージや単球に対して、食作用や走化性の増加、腫瘍細胞への細胞毒性の増加などの影響を与える[8]。M-CSFの役割は、単球/マクロファージ細胞系に限定されるものではない。M-CSFは、その膜受容体(c-fmsがん原遺伝子によってコードされるCSF1R(英語版)またはM-CSF-R)と相互作用することにより、初期の造血前駆細胞の増殖を調節し、免疫学、代謝、生殖、妊娠に関わる多くの生理学的プロセスに影響を与える[9]。
また、副甲状腺ホルモンによる内分泌刺激を受けた骨芽細胞から放出されるM-CSFは、破骨細胞に対して傍分泌作用を発揮する[10]。M-CSFは破骨細胞の受容体に結合して分化を誘導し、最終的には骨の再吸収(英語版)(破壊)を通じて血漿カルシウム濃度の上昇をもたらす[要出典]。また、妊娠中の子宮内膜上皮にはCSF-1が多く発現しており、胎盤の栄養膜(英語版)にはCSF-1の受容体であるCSF1Rが多く発現していることが確認されている。これまでの研究で、局所的な高濃度のCSF-1が栄養膜のCSF1Rを活性化することが、正常な胚の着床と胎盤の発育に不可欠であると明らかになっている。さらに最近では、CSF-1とその受容体であるCSF1Rが乳腺の正常な発育と腫瘍化に関与していることが明らかにされた[11]。
臨床的意義
局所的に血管壁で産生されたM-CSFは、アテローム性動脈硬化症の発症と進行に寄与する[12]。
また、M-CSFは、急性腎障害や慢性腎不全などの腎疾患にも関与していると言われている[13][14]。単球の慢性的な活性化は、慢性腎不全患者の複数の代謝的、血液学的、免疫学的な異常を引き起こす可能性がある[13]。急性腎障害では、M-CSFは損傷後の修復を促進することが示唆されているが[15]、反対に炎症性マクロファージの表現型を増殖させることも報告されている[16]。
薬剤の標的
PD-0360324(英語版)やMCS110(英語版)は、幾つかのがんを対象に臨床試験が行われているCSF1阻害薬である[17]。
相互作用
M-CSFはPIK3R2(英語版)と相互作用する[18]。
